CRISPR PATTERN
CRISPR/Cas9 is the most widely used method for genetic manipulation today. Originally, bacteria used the CRISPR system as an adaptive and inheritable immune system for the defence against viral DNA and plasmids. They snip out parts of the viral DNA and store them to recognise the pathogen if they have to face it again.
In 2012, Dr Jennifer Doudna and Emmanuelle Charpentier successfully adapted CRISPR so that it could be used in animal and human cells. This breakthrough created a buzz in the scientific world and revolutionised gene editing. To this day, CRISPR is the simplest and most precise genome engineering technique available.
CRISPR uses a guide RNA (gRNA) that is designed to find and bind to a specific sequence in the DNA. This gRNA guides the Cas9 enzyme to its target location. Here it acts as a pair of molecular scissors that cuts both DNA strands good at a specific location. This initiates the cell's own DNA damage repair mechanism, which introduces changes to the genetic code, changing the composition of expressed proteins.
